Clonal propagation of roselle (Hibiscus sabdariffa L.) by shoot tip culture for salinity tolerance

Abstract:

Actively growing shoot apices obtained from selected 35 days old roselle (cv. Aswan ) plants were used in this study. Murashige and Skoog medium was used. Salts of potassium nitrate, potassium phosphate and magnesium sulphate with three concentrations for each salt were studied. In vitro experiment results showed that number of shoots, roots, leaves/plantlet and shoot length of plantlet mean values werehighest under the treatment of 400, 40 and 1.6 mg/l for the three previous salts, respectively. In a greenhouse, plants tolerated the irrigation water salinity up to 7000 mg/l, and plant height was 66.19 cm and number of flowers/plant 54.26 and fresh and dry weights of calyces were79.92 and 10.03 g/plant, respectively.
