Slow-growth preservation of wild shih (Artemisia herba-alba Asso.) microshoots from Jordan

Abstract:

The biodiversity of shih (Artemisia herba-alba) is heavily subjected to loss because of heavy grazing, land cultivation and collection by people to be used in folk medicine. Slow growth in vitro preservation using different osmotic agents (sucrose, sorbitol and mannitol) with different concentrations (0.0, 0.1, 0.2, 0.3 or 0.4 M) or abscisic acid at different concentrations (0.0, 0.5, 1.0, 1.5 or 2.0 mg.l-1) was studied. Preservation of the microshoots on a medium supplemented with 0.1, 0.2 or 0.3 M sucrose, mannitol or sorbitol under light at 25°C was able to inhibit the growth rate and maintain explant quality up to 12 weeks. Using ABA at concentration 0.5 mg.l-1 reduced growth and maintained complete survival and complete regrwoth ability after preservation for 12 weeks.
