Isolation of denitrifying bacteria from groundwater in typical intensive vegetable cultivation area

Abstract:

The Northern China Plain produces over 20% of the national grain production. Land management practices affect the nitrogen fate and availability of NO3--N, increasing leaching in the main pathways that impact groundwater quality. Experimental Research Station was contaminated with nitrate due to large nitrogen fertilizer applications. To remediate the polluted groundwater, we carried out to get effective denitrifying bacteria. A denitrifying bacteria strain (named VOTO1-F) with 60.5% denitrifying intensity was isolated from the intensive vegetable production area in Shouguang, China. Strain VOTO1-F, identified as a strain of Agrobacterium sp. by plate serial dilutions, 16S rDNA gene amplification and denaturing gradient gel electrophoresis (DGGE), is expected to be of a source organism for in situ bioremediation of groundwater contaminated nitrate.
