Using of random amplification of polymorphic DNA (RAPD) analysis for typing of different Escherichia coli isolates

Abstract:Application of random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) as a tool for molecular typing of different Escherichia coli isolates was studied in order to ensure the discriminatory power of this technique among Escherichia coli strains isolated from bovine fecal and milk samples. Traditional typing techniques based on phenotypic characteristics such as serotyping are being increasingly challenged by the use of DNA-based methods. A set of five primers was evaluated for their ability to produce a high number of distinct fragments. Isolates of E. coli typed by random amplification of polymorphic DNA (RAPD) revealing 2 clusters of strains were defined on the basis of the epidemiological data and subsequently phylogenetic tree was created. The obtained results indicated that the serotypes causing bovine mastitis were genetically similar to that causing diarrhea.
