Determination of biogenic amines in cheese by nanofiber solid phase extraction-reversed phase high performance liquid chromatography with pre-column derivatization

Abstract:

A novel analytical method with nanofiber solid phase extraction (NF-SPE) has been developed for the simultaneous determination of histamine (Him), tyramine (Tyr), putrescine (Put), cadaverine (Cad) and tryptamine (Try). The separation of five biogenic amines (BAs) was carried out by using a high performance liquid chromatography (HPLC) on a reversed-phase column (Agilent Hypersil ODS column, 125 mm×4.6 mm×5 µm) with pre-column derivatization and fluorometric detection (FLD, excitation wavelength at 340 nm and emission wavelength at 450 nm), and the elution buffers were based on sodium acetate with the flow rate 1.0 mL/min. The five BAs could be separated within 15 min. All the BAs exhibited good linearity over the tested range (0.01-25.0 mg/L); least square analysis produced a correlation coefficient between 0.9970 (Try) and 0.9997 (Him). The limit of quantitative analysis was estimated to be varying between 0.005 mg/L (Him, Tyr) and 0.01 mg/L (Put, Cad, Try) under trichloroacetic acid extraction and NF-SPE process based on the calibration curves and recovery rates between 92.6% (Cad) and 107.4% (Tyr). The repeatability tests of HPLC instrument and NF-SPE preparation for samples were performed respectively during a working day, expressed as R.S.D., ranged from 0.58% to 4.32% (repeatability of HPLC instrument), from 2.00% to 6.01% (repeatability of NF-SPE preparation). Comparison of five BAs analysis by the direct extraction method (no NF-SPE process ) and the proposed extraction method (including NF-SPE process) showed preliminary NF-SPE cleaning process that can eliminate the impurity that coeluted with BAs in the direct extraction method. The presented method is simple, fast, and reliable, and can be applied for the quantitation of five BAs in cheese samples efficiently after extracting with 50 g/L trichloroacetic acid and purified with NF-SPE method.
