Effect of storage methods on virulence variation of Xanthomonas oryzae pv. oryze

Abstract:

Eight representative isolates of Xanthomonas oryzae pv. oryzae from Philippines were maintained using five kinds of storage methods for 3-5 years while their virulence was tested on five differential varieties every year. The results showed that some isolates were able to attenuate their virulence under any storage condition while the good storage methods did keep the virulence of the isolates for longer period of time without any remarkable changes. In particular, lyophilization almost maintained the virulence of all the isolates after 5 years storage. The results also showed that some isolates were stable in their virulence regardless of the storage method. Overall, this result indicated that the lyophilization and deeper freezer storage should be recommended in the identification of the strains, while it is necessary to use the isolates with stable virulence in screening for the resistance.
