Characterisation of CDX2 in grass carp (Ctenopharyngodon idella) and its regulation by diets

Abstract:

The CDX2 gene is a crucial factor which regulates intestinal cell proliferation and terminal differentiation and abundantly expressed in intestine. In this study, we cloned grass carp the CDX2 sequences and characterised its expression profile upon dietary protein. The CDX2 sequence encodes a 258 amino acids which shows a high similarity among vertebrates. Expression analysis revealed that the CDX2 of grass carp showed the highest expression in the intestine with no significant difference with longitudinal distribution. During embryonic development, CDX2 showed a dynamic pattern of expression during development. It reached the maximum in the hatching stage and minimum in the blastocyst stage. After hatching, the CDX2 expression showed constant levels from 42 and 49 day in fish fries. To determine whether fish diet may have any effect on CDX2 expression, we extended our research to dietary regulation of CDX2. The CDX2 mRNA level was low in grass carp fed with 27%, 32% and 42% CP, while the highest expression was found in grass carps fed the 37% CP diet (P < 0.05) in 7, 14 and 21 days. In addition, CDX2 mRNA levels were higher after fed with soybean meal than fish meal and mixture meal. Together, the results demonstrated for the first time that CDX2 mRNA expression is regulated in a temporal and spatial pattern during development, and dietary protein had significant effects on CDX2 expression in grass carp.
