Fluorescein diacetate and propidium iodide FDA-PI double staining detect the viability of Microcystis sp. after ultrasonic irradiation

Abstract:

This paper studied the Microcystis sp. cells viability tests by the method of double staining with fluorescein diacetate and propidium iodide (FDA-PI), and the impact on Microcystis sp. cell viability under different ultrasound intensity. Living cells were bright green and dead cells were red after FDA-PI double staining, so the method can distinguish dead cells and living cells intuitively. Research also showed that cells were green and dark red at lower ultrasound intensity (below the cavitation threshold), whereas cells mostly were dark red and sporadic bright green at high intensity (above the cavitation threshold). Most Microcystis sp. cells were damaged and cell viability decreased at low intensity, while most cells died after high intensity ultrasound treated.
