A standardized protocol for the genomic DNA isolation from the leaf of Camellia sinensis (Linn.) O. Kuntze

Abstract:

Relatively high amount of DNA could be extracted from the leaf sample of Camellia sinensis (Linn.) O. Kuntze by following the standardized and modified protocol. Isolated DNA was quantified using a spectrophotometer at the absorbance of λ260  and λ280 nm. Tea being an important commercial crop in India, many research works has been done to improve tea quality and yield. Clonal varieties have been developed which have considerable genetic variability. Eight clonal varieties of Camellia sinensis (Linn.) O. Kuntze were selected from various tea gardens of Golaghat district, one of the major tea producing districts of Assam, India.  The collected clonal varieties were all produced in the Tocklai Tea Research Institute, Assam. All the clonal varieties yielded a good quality DNA in a range of 1.81-2.77 mg/ml  and produced clear bands on the agarose gel. The estimation of purity of the obtained DNA was concluded by analyzing the ratio of λ260 nm/λ280 nm of the genomic DNA, which ranged 1.93-2.73. The obtained DNA was then statistically analyzed for concentration and purity, which showed considerable differences.
