Purification and partial bioactivity in vitro of polysaccharides from sporophyll of Undaria pinnatifida

Abstract:In China, sporophyll of brown seaweeds, Undaria pinnatifida, as processing waste was discarded, that caused a great waste of resource and induced environment pollution. Polysaccharide from the sporophyll of Undaria pinnatifida (PSU) is interesting due to its various biological activities. This polysaccharide was isolated from raw material by dilute acid extraction and crude extract was purified by chromatography on DEAE-cellulose. Four fractions of PSU were obtained and the composition of the four fractions of PSU was characterized by organic elemental analysis and GC. Comparing the other fractions, PSU I contained the most protein and uronic acid and PSU III the most carbohydrate and sulfate. Fucose, galactose and mannose were contained in all of fractions, but rhamnose and glucose were contained only in PSU I. The ratio of galactose and fucose in PSU IV (0.88:1) and PSU III (0.32:1) was higher than in the other fractions, which was the critical factor to its anti-inflamatory and antioxidant activities. Biological activity research in vitro showed that PSU IV could inhibit RAW 264.7 cell, releasing NO induced by LPS. PSU IV had a strong resistance to cell oxidative damage caused by tBOOH and all of the fractions could remarkably promote lymphocyte proliferation.
