Quantitative determination of polyphenols in tobacco leaves by HPLC

Abstract:

A high performance liquid chromatographic (HPLC) method for the determination of 9 polyphenols in tobacco was studied. These polyphenols were extracted from tobacco sample by ultrasonic extraction method. Polyphenols were separated on a Waters Sunfire C18 (4.6 mm ×150 mm, 5 µm) chromatographic column. The mobile phase was methanol, water and 2% HAC solution on gradient elution at a flow rate of 1 ml/min. Each polyphenol was monitored by UV detector and 9 polyphenols in tobacco could be simultaneously determined by this method. The recovery of these polyphenols was 94~ 99%, and relative standard deviation from 0.44% to 4.26%. This method was applied to the determination of polyphenols in tobacco with good results.
