Embryo rescue to obtain progeny from seedless grapes (Vitis vinifera L.) × wild Chinese Vitis species

Abstract:

For the purpose of developing an efficient protocol for hybrid embryo rescue of seedless grapes × wild Chinese Vitis species and obtaining progenies from seedless grapes (Vitis vinifera L.) × wild Chinese Vitis species, two seedless grape cultivars (Emerald seedless and Crimson seedless) as female parents and two wild Chinese Vitis species of Yanshan (V. yanshanensis J. X. Chen) and Jiangxi-2 (V. romanetii Roman.) as male parents were used to make four hybrid combinations. The effects of sampling time, plant growth regulators and parent genotypes were investigated to improve embryo rescue technology in this study. The results indicated the best sampling time for ovule inoculation differed from different cultivars: 47 days after pollination for Emerald seedless and 60 days for Crimson seedless. These in vitro ovules were cultured on ER medium, and then all embryos developed were excised from ovules and cultured on 7 different media (WPM with different plant growth regulators). The highest rates of embryo germination (83.3%) and plant development (70.0%) were found when embryos were cultured on WPM + indole-3-acetic acid (IAA) 1.0 mg·l-1 + 6-benzyladenine (6-BA) 1.0 mg·l-1, supplement with 20 g·l-1 sucrose, 500 mg·l-1 casein hydrolysate (CH), 3 g·l-1 activated charcoal (AC) and 7 g·l -1 agar. These 4 hybrid combinations were examined and the production of hybrid plants was significantly different among different genotypes, ranging from 70.2% (Emerald seedless × Yanshan) to 51.0% (Crimson seedless × Jiangxi-2). A total of 374 seedlings were acclimated and transplanted to the field in 2011 and 2012. Therefore, this embryo rescue method has been used successfully to produce novel hybrids that may be useful to improve disease resistance of seedless grape cultivars.
