Detection of Elsinoë fawcettii in planta through real-time PCR

Abstract:

Citrus scab, caused by Elsinoë fawcettii, is one of the major diseases in citrus worldwide. Here we present a PCR-based technology for detection of E. fawcettii in planta. The primers used in this technology were designed specifically for the nucleotide sequence within the ITS region of E. fawcettii genomic DNA. Using this primer set, we were able to amplify a 275-bp fragment from all the E. fawcettii isolates collected from citrus grown in different geographic locations. No specific products were amplified from DNAs of other four different fungal pathogens that are commonly present on citrus leaves using these primers. This newly developed real-time PCR can also be used to detect early infection of E. fawcettii in citrus leaves efficiently, and thus may help field pathologists and citrus growers to decide the time of fungicide applications.
