Optimization of ultrasonic-assisted extraction of total flavonoids from Petasites tatewakianus using response surface methodology

Abstract:

Due to their widespread usage as herbal supplements for anti-spasmodic and anti-inflammatory effects, different species of Petasites are of interest to scientists of various disciplines including food, health, pharmacology, chemistry, botany and toxicology. Response surface methodology (RSM) was applied to plant extracts of P. tatewakianus for the parameter-optimization of ultrasonic-assisted flavonoid extraction. The effects of extraction temperature, the liquid to solid ratio, ethanol concentration and extraction time, on the yield of RSM-based flavonoid extraction were investigated. Statistical analysis show ed that the extraction time, ethanol concentration, and the quadratics of the selected four variables showed significant effects on total flavonoid yield. The optimal extraction condition was at 70.63°C, 68.19% ethanol, with the liquid to solid ratio of 60.57:1 (ml/g) for 2.67 h, and the predicted total flavonoid yield was 14.31mg/g under this condition. The optimal conditions could be modified as 71°C, 68% ethanol, with the ratio of liquid to material of 61:1 for 2.67 h (160 min). Under the modified conditions, the experimental yield of flavonoids was 14.11± 0.20 mg/g.
